Effects of bisphenol A exposure during cardiac cell differentiation

چکیده

Heart development requires a precise temporal regulation of gene expression in cardiomyoblasts. Therefore, the transcriptional changes differentiating cells can lead to congenital heart diseases. Although genetic mutations underlie most these alterations, exposure environmental contaminants, such as bisphenol A (BPA), has been recently considered risk factor well. In this study we investigated genotoxic and epigenotoxic effects BPA throughout cardiomyocyte differentiation. H9c2 (rat myoblasts) were exposed 10 30 ?M before during last two days cardiac-driven Then, have analysed phenotypic molecular modifications (at transcriptional, epigenetic level). The results showed that treated myoblasts developed skeletal muscle cell-like phenotype. induced by genes codifying proteins involved differentiation function depend on window BPA. repressed transcription factors (Hand2 Gata4), whereas reduced cardiac-specific (Tnnt2, Myom2, Sln, Atp2a1). Additionally, significant observed regarding DNA damage histone acetylation levels after periods exposure: toxicant percentage repair foci (formed co-localization ?H2AX 53BP1) increased dose-dependent manner, treatment with triggered decrease marks H3K9ac H3K27ac. Our vitro reveal seriously interferes process differentiation, which could be related reported vivo cardiogenesis.

Human Peripheral Blood Mononuclear Cell Function and Dendritic Cell Differentiation Are Affected by Bisphenol-A Exposure

Environmental pollutants, including endocrine disruptor chemicals (EDCs), interfere on human health, leading to hormonal, immune and metabolic perturbations. Bisphenol-A (BPA), a main component of polycarbonate plastics, has been receiving increased attention due to its worldwide distribution with a large exposure. In humans, BPA, for its estrogenic activity, may have a role in autoimmunity, in...

Immunoregulatory Effects of Glutathione During Mesenchymal Stem Cell Differentiation to Hepatocyte-Like Cells

Background: The role of mesenchymal stem cell in cellular therapy is the subject of interest for many researchers. The differentiation potential of MSCs and abilities in modulations of the recipient’s immune system makes them important cells in tissue regenerative studies. MSCs by releasing the proinflammatory cytokines play important role in immunomodulatory systems; however the signaling path...

Adverse Effects of Bisphenol A Exposure on Glucose Metabolism Regulation

Bisphenol A (BPA) is used as basic chemical compound in the production of polycarbonate food containers or epoxy resins coating metallic cans for food and beverages conservation. Its xeno-estrogenic activity alters endocrine-metabolic pathways modulating glucose metabolism and increasing the risk of developing diabetes, insulin resistance, and obesity. Based on in vitro and in vivo experimental...

Bisphenol A exposure, effects, and policy: a wildlife perspective.

Thousands of anthropogenic chemicals are present in the environment, and mounting evidence indicates that some have endocrine-disrupting effects in a variety of organisms. Of particular concern are chemicals that act as agonists or antagonists on vertebrate estrogen or androgen receptors. One such compound is bisphenol A (BPA), which appears to be both an estrogen receptor agonist and an androg...

The Genotoxic and Cytotoxic Effects of Bisphenol-A (BPA) in MCF-7 Cell Line and Amniocytes

Bisphenol-A (BPA) is an industrial xenoestrogen used widely in our living environment. Recently, several studies suggested that BPA has destructive effects on DNA and chromosomes in normal body cells via estrogen receptors (ER). Therefore, BPA could be considered as an important mediator in many diseases such as cancer. However, there are still many controversial issues which need clarification...
